Since esperanto has the second person as both singular and plural (like in english), if I wanted to express a more definite sense of plurality, could i say, "Vi du estas virinoj." -- "You two are women." expressing that in a more subject like way than "Vi estas du virinoj."

Do the way you like. Both vi du estas inoj and vi estas du inoj are good. But actually for such phrases there're collective numbers in esperanto formed by a suffix -op-, which, to my mind, are more preferable, so you can use duopo or ambaŭ: vi duopaj estas inoj, vi duopo estas inoj, vi ambaŭ estas inoj.
However, in fact, esperanto doesn't have an exact way you must say things, any linguistic stereotypes or clichés. Say the way you want and enjoy it.
