Specialiĝo sounds ok at least according to Reta-Vortaro.
Mi specialiĝas en lingvistikon en/ĉe universitato.
The English word "college" is tricky one, because it means different things in different countries. If it means the level after elementary school, use "mezaltlernejo". If university use "universitato".
Mia specialiĝo/specialiĝa fako en la mezaltlernejo/universtato estas malfacila.
Possibly you can leave out the educational institution, if the listener already knows it. Thus
Mia specialiĝo/specialiĝa fako estas malfacila.
