I need help, i need to know the difference between both.

Adjectives or a-vortoj can only be used to describe nouns.
 
  • La plej bona aŭto estas tia, kiu… : The best car is such that…
  • El tiuj aŭtoj mia aŭto estas la plej bona. : Of those cars my car is the best one.
In both sentences bona refers to a car (bolded).

If the thing you are referring to is something else than a noun, you use adverbs or e-vortoj (n.b. adverbs in English have a narrower field of use).
 
  • La plej bone vendata biero en Domingo estas… : The best selling beer in Dominican Republic is…
where bone refers to an adjective, "vendata" (bolded).

Note following uses of adverb
 
  • Estas bone vidi vin. (there is no subject) : It is good to see you ("it" is a so called formal subject required by the English grammar)
  • Mi fartas bone. (refers to a verb) : I am doing well.
  • Nenie estas bone! (refers to a correlative) : Nowhere is good!

As Metsis points out 'la plej bona' is used when reference is made to a noun (or pronoun) . The reference may be explicitly stated or understood.

Although there are examples in the Tekstaro of Zamenhof using la plej bone to qualify a verb This does not seem to have widely copied by later authors.

Personally I would say ekz. Li plej bone konis la spiriton de Esperanto, rather than Li la plej bone konis la spiriton de Esperanto.
