There's a big difference between (say) running in the room and running into the room. Since Esperanto offers only "en" for both "in" and "into," I would like to know how the two meanings are distinguished.

I have a vague recollection that "en la ĉambro" means "in the room" and "en la ĉambron" means "into the room," but I can find no mention of that in the Grammar that's part of this site.

Your recollection is correct, see section Akuzativo, chapter Direkto.

Multaj dankojn.

La votaro de Lernu diras: "en (en) - in, within, inside, into, on, per." Tio estas kial la demando okazis.

The dictionary of Lernu is a good starting point, use it to get a rough translation, but check against some more trustworthy sources (see my recent posting here in Lernu).
