I want to say something like
"I am back here in five minutes".
How to say this in Esperanto?
"Mi returnas en kvin minutojn"?
Is "en" the correct preposition?

If you want to say that you'll return when 5 minutes have past, it would be "post". If you just want to show that you'll return at some point within 5 minutes, say "en".

It may be instructive to study the sections of PMEG concerning the different prepositions dealing with time:
en (section "Tempo"), "within".
dum, "during".
tra (section "Tempo"), "through".
por (section "Tempo"), "for" (in the sense of a desired period).
ĝis, until (an event has occurred, not usually the lapse of a measured period of time)
antaŭ (section "Tempo"), "before", but note also the use of antaŭ ol in various circumstances.
post (section "Tempo"), "after", but note also the use of post kiam.

Yes, for example you could say Tio bezonos inter kvin kaj dek minutojn, "That will take between five and ten minutes", or Tio bezonos ĉirkaŭ sep minutojn, "That will take about seven minutes". There are examples in PMEG for inter (section tempo) and ĉirkaŭ (section Proksimumeco).

"Inter" is a preposition. The accusative does not follow a preposition unless direction is indicated.

Generally that's true, but it's not an absolute and is not the case here. PMEG has the following quote:

Por aĉeti ĝin vi devas pagi inter 100 kaj 200 eŭrojn.
