In my opinion, anta is correct here. I almost agree with Dmitrij 'Esprimmaniero' except for the second la. I would put it la tria plej rapide kreskanta regiono de la lando.

Good question; I would say no, on the grounds that since tria refers to the phrase plej rapide kreskanta taken as a whole to be an adjective, under such circumstances the adjectival form, not the adverbial form, is used. See the last example in the third box in the following
page from PMEG.

"la trie plej rapide kreskanta lando" (or "nacio") would mean "the land/nation which is, third of all, the fastest growing" (while first of all, it is the tiniest, second of all it is the warmest, etc). I use "unue, due, trie, ..." when listing points. "La dua plej alta domo en la urbo" is acceptable and correct, and therefore I think your form is also correct the way it is.
