Would 'preni' have the meaning of taking as in a road, or would some other words be more correct?

I think something like 'eklaŭiri' would certainly be more expressive.

I would avoid “preni.” “Elekti” might work. “Ekveturi” might give more of the meaning.

I've seen it used, and I don't think preni vojon would confuse anyone.

I concur. The following example given in the Reta Vortaro entry for preni seems to suggest the root is ok:

Reta Vortaro:ekpreni la vojon al iu loko;

I might use elekti or iri laŭ, but, as others have indicated, if you said Mi prenis la dekstran vojon, you would probably be understood.
